It has previously been shown that antibodies against the transmembrane proteins, band 3 and glycophorin A, inhibit entry of the merozoite into the red cell and, in the case of band 3, it was established that attachment of the parasite to the cell is not prevented. We have found that antibodies against the cytoplasmic domains of band 3 and of glycophorin A, when present in the interior of resealed ghosts of human red cells, also inhibit invasion by P. falciparum. It is inferred that attachment of the merozoite to the red cell causes structural effects that are transduced to the membrane cytoskeleton and the antibodies against transmembrane proteins interfere with the invasion sequence at this level.

The life-cycle of a precocious and attenuated line (WisF96) of Eimeria tenella, derived from the Wisconsin (Wis) strain, contained only the first of the three generations of schizogony undergone by the parent strain. The reproductive capacity of WisF96 was less than that of the parent strain by a factor of about 2000, but inoculation of chickens on two occasions with a large number of its oocysts induced resistance against challenge with oocysts, or with second-generation merozoites of Wis. The immunizing abilities of the attenuated line and its parent were compared by priming groups of chickens with numbers of oocysts of WisF96 or Wis, designed to produce infections of equal magnitude in terms of oocyst production (standard inocula), and then challenging with oocysts of Wis. The results suggested that an equivalent parasite mass of WisF96 was more immunogenic than Wis. This was confirmed by the finding that, even when the priming dose of Wis was increased 10-fold, the standard inoculum of WisF96 still produced the greater protective effect. These results indicate that the first generation of schizogony of E. tenella is highly immunogenic.

During my third year as an undergraduate reading Zoology at Cambridge, I found a relatively large parasite inside one of the Asellus aquaticus that had been allocated to me for the afternoon's practical work. After we had failed to identify even the group of animals to which it belonged, someone suggested that I should take my find to The Molteno Institute on the Downing Site. There I was ushered into Room 3 where I first met Dr Tate. After listening to my account he looked at my specimen and said, ‘Now just one second …’ in his rich Irish accent. As I later came to know, so much of his teaching or advice began with that highly characteristic phrase. He told me that I had found an acanthocephalan worm in its intermediate host, and when I returned some days later to tell him the results of my search of the literature, he suggested that I should work on the Acanthocephala under his supervision for the Ph.D. Degree after my graduation. I never regretted for one moment my decision to take up his offer. He maintained close contact with me, as he did with all his former research students, and the last letter I received from him was dated 16 September 1985, written just a few weeks before his death on 7 November 1985 in hospital in Cork. During the 24 years of our association I was never on Christian-name terms with him. He was always ‘Dr Tate’ or, when talking with equally close mutual associates like Donald Lee, David Molyneux, John Barrett, Vaughan Southgate or Roger Tatchell, we referred to him as ‘P.T.’. He had great integrity and fairness in all that he did and an ability of seeming to know exactly what you were thinking. These attributes, coupled with his extraordinarily wide parasitological knowledge, generated our respect. He also had a delightful sense of fun and even his sarcasm was kind and gentle. Our children loved him and his steady supply of sweets for those who visited him in Room 3 and the book tokens that came without fail every Christmas were expressions of his kindness.

Changes in the response of black mollies Poecilia latipinna to infection with Ichthyophthirius multifiliis subsequent to single I. multifiliis infections were examined experimentally. Incomplete resistance to infection was established, the degree of which did not depend on the intensity of the initial infection. Resistance was maintained for longer periods, however, by those fish with higher initial levels of infection. Previous experience of infection by their hosts had a significant, though small, effect on the time for which trophozoites remained on the fish. The influence of these results on the population dynamics of the Ichthyophthirius-fish interaction is examined with the aid of simple mathematical models.

The maintenance of Theileria parva parva infection in an endemic area of Kenya on the shore of Lake Victoria was studied in the field and laboratory. High prevalences of antibodies against T. parva and T. mutans and intra-erythrocytic piroplasms were detected in local zebu (Bos indicus) cattle. The mean infection rate of Theileria parasites in the tick, Rhipicephalus appendiculatus, in field collections was 1·1 %. Most of the infection was attributed to T. parva parva by application of field ticks to susceptible cattle. Five cattle, all about 1·5 years old, were purchased from local owners and transported to the laboratory. All five had oscillating antibody titres against T. parva and T. mutans and had patent theilerial infections during the subsequent 13 months. Uninfected R. appendiculatus nymphs were applied to cattle at 0, 3, 6, 9 and 13 months after transport to Muguga, and 18 out of 23 batches transmitted T. parva parva infection to cattle when 100 resultant R. appendiculatus adults were applied. Infection rates in the tick batches were usually low, with 1 salivary gland acinus infected/tick. Hence, a frequent carrier state of naturally infected cattle has been demonstrated for T. parva parva for the first time, and it is likely that this carrier state is of great importance in maintenance of T. parva parva infection in the field.

Scanning electron microscopical studies of adult male and female Schistosoma curassoni, male S. haematobium and male S. bovis are reported. The tubercles on the dorsal and dorso-lateral surfaces of male S. bovis are devoid of spines, whereas spines are present on the tubercles of male S. curassoni and S. haematobium. Some of the tubercles of S. curassoni and S. haematobium are completely covered in spines, whereas others have a spine-free apical area. Thus, on the isolates examined from Senegal it is concluded that adult male S. bovis can be easily distinguished from S. curassoni or S. haematobium on the basis of the structure of the tubercles. The surface of female S. curassoni is smooth, with transverse striations and a pitted appearance, apart from the posterior part where anteriorly pointing spines are found.

The distribution of anionic residues on the surface of erythrocytes infected with Plasmodium falciparum was studied using cationized ferritin (CF) and transmission electron microscopy. CF staining of uninfected erythrocytes or erythrocytes infected with a knobless variant resulted in a dense and uniform distribution of ferritin particles; however, when red cells infected with a knob-inducing variant were exposed to CF, aggregates of ferritin particles were observed in the region of membrane elevation. Lectin binding to the erythrocyte surface was visualized by transmission electron microscopy using ferritin-conjugated lectins and lectin-fetuin-gold. No differences were observed in the lectin-binding patterns of malaria-infected or uninfected erythrocytes using WGA (wheat-germ agglutinin), RCA (ricin), and Limax flavus lectin. In distinct contrast to the uniform distribution of ferritin particles seen with these lectins was the appearance of clusters of ferritin-ConA over the knobby regions. Localized aggregates of ConA were not seen in knob-free areas or on the surface of red cells infected with a knobless variant. No significant differences were found in the agglutination reactions of normal and infected cells with the Cancer antennarius lectin specific for O-acylated sialic acids.

Biomphalaria glabrata snails are known to be castrated by infection with the trematode parasite Schistosoma mansoni 4–6 weeks post-infection. The pattern of oviposition in the first 35 days post-exposure (p.e.) was investigated, in snails aged 14 weeks and measuring 7–10 mm diameter which had not commenced egg-laying, by counting the numbers of eggs laid in 7-day intervals. A group of exposed snails was compared with a control non-exposed group. The exposed group included both parasitized and non-parasitized snails, and showed a significant increase in the median number of eggs laid during the periods 14–21 and 22–28 days p.e. Throughout the entire 35-day period exposed non-parasitized snails laid significantly more eggs than control snails, while parasitized snails laid significantly more eggs than controls during days 22–28 p.e. and significantly fewer during days 29–35 p.e. Parasitized snails also laid significantly more eggs/egg mass in the period 16–28 days p.e. than did control snails. Growth of the snails was measured. By day 28 p.e. the mean diameter of the exposed group was significantly greater than that of the control group. The increase in oviposition by snails soon after exposure is discussed in terms of a compensatory response for expected future suppression of egg-laying. The fact that parasitized and non-parasitized snails both show increased oviposition indicates that normal development of the parasite is not necessary to trigger the response.

An automated suspension culture system for the cultivation of Plasmodium falciparum is described which retains a degree of flexibility which is absent in other automated culture apparatuses. Not only does this system of cultivation promote rapid multiplication of asexual parasites but also permits the development and maturation of gametocytes. Using a combination of gelatin flotation and N-acetyl glucosamine treatment synchronous development of gametocytes was achieved. The total time for gametocyte maturation in vitro under the conditions provided was 7 days. Stages II and V required 48 h for development whilst I, III and IV needed 24 h each. Mature microgametocytes were relatively long lived in comparison with macrogametocytes. Electron microscopic study of the synchronized stages confirmed the observations of Sinden (1982) but, in addition, we noted the presence of Garnham bodies, a cytostome in all stages and dense spherules in stages I–III similar to the fenestrated buttons in sporozoites and exoerythrocytic forms. The relationship between the number of osmiophilic bodies in the mature gametocytes and their ability to escape from the red cell is reaffirmed.

Adults of Pseudodiplorchis americanus occur in the host's urinary bladder, the typical habitat of polystomatid monogeneans in amphibians. However, larvae which invade the respiratory tract via the nostrils then migrate to the bladder through the stomach and intestine, a route which is without precedent amongst monogeneans. Parasite transmission is correlated with the spawning of the desert toad Scaphiopus couchii (June/July in Arizona, USA) and the gut migration, which takes as little as 5 min, occurs after 26 days post-infection (p.i.). Migration occurs during the host's feeding season, and therefore the parasites normally encounter maximum digestive activity along their path. Worms transferred experimentally from respiratory tract to the digestive fluids in the gut are killed within 60 s. However, natural migration is stimulated by an unidentified host factor which triggers protective adaptations, enabling parasite survival for up to 4 h in these lethal conditions. Without this essential stimulus, migration may be delayed for over 1 year and juveniles remaining in the respiratory tract undergo no development beyond that reached at 4 weeks p.i. Rapid reproductive development is resumed immediately after migration, leading to maturation around 1 month later and progressive accumulation of embryos in utero in preparation for transmission during the next summer's rains.

Sporozoites of Plasmodium yoelii yoelii were incubated for 40 min with BALB/c peritoneal macrophages in the presence of nitro-blue tetrazolium (NBT). While immature oocyst sporozoites triggered the macrophage respiratory burst, as visualized microscopically by the localized reduction of NBT to insoluble formazan, 97·6% of mature salivary gland sporozoites did not induce such a response. The macrophage oxidative response was also induced by 82·7% of heat-inactivated and 95·7% of trypsin-treated salivary gland sporozoites. The relationship of these results to the infectivity and immunogenicity of malarial sporozoites is discussed.

The uptake of radioactive pyrimethamine by a sensitive and a resistant strain of Plasmodium falciparum, the metabolic fate of pyrimethamine inside these parasites and the kinetic properties of dihydrofolate reductase (DHFR) from both strains have been studied. Uptake of the drug was identical in both strains and no metabolite of pyrimethamine was found in either strain. DHFR from the resistant strain was 300 times less sensitive to inhibition by pyrimethamine than the enzyme from the sensitive strain, while the Michaelis constant for dihydrofolate remained unchanged and inhibition was competitive in both cases. Altered properties of plasmodial DHFR are apparently the only mechanism responsible for pyrimethamine resistance in the strain of Plasmodium falciparum studied.

Four common blood parasites belonging to the genera Babesia, Hepatozoon, Trypanosoma and Grahamella were detected in sympatric populations of Clethrionomys glareolus and Apodemus sylvaticus. Seasonal changes in the abundance of hosts and parasites were monitored in a longitudinal field survey for 18 months. All parasites in both hosts showed similar seasonal patterns with most transmission occurring in summer and autumn and ceasing in winter. Amongst aged subsets of each host population, Hepatozoon and Grahamella spp. were primarily parasites of young animals whereas Babesia and Trypanosoma spp. generally infected older hosts. Data from the aged subsets were also used to estimate values for the basic reproductive rate, R0, for each parasite. In C. glareolus, R0 = 2·0 for B. microti, 2·6 for H. erhardovae, 1·7 for T. evotomys and 2·8 for Grahamella sp. In A. sylvaticusR0 = 2·2 for Grahamella sp. These estimates are probably representative only for summer and autumn of each year.

An enzymatic comparison has been made between isolates of Schistosoma mansoni from rats and humans in Guadeloupe and a Burundi isolate of S. rodhaini. Analyses of LDH, MDH, AcP, PGM, GPI, G6PDH and HK by isoelectric focusing provided no evidence for the involvement of S. rodhaini in the recent evolution of the schistosomes currently endemic in Guadeloupe. No distinction could be made between murine and human isolates of S. mansoni and it is suggested that murine schistosomiasis should not therefore be ignored in control programmes. Rattus rattus were captured at seven sites around the island; of 142 examined, 48 were positive for schistosomes. Differences in prevalence between habitats were marked and only small changes in prevalence were observed in localities sampled in 1982 and 1983. Animals with the greatest worm burdens were associated with areas of high prevalence, and age-related changes in worm burden were observed. Two alleles, a and b, at the MDH-1 locus of S. mansoni from rats were identified. Differences in the overall frequencies of these alleles were observed for schistosomes from different localities. Allelic frequences representative of schistosomes from rats at four localities were stable from 1982 to 1983. The majority of positive animals, even those with light worm burdens, were found to be infected with a number of different schistosome genotypes.

The structure of the ovarian tissue in juvenile Corynosoma semerme and C. strumosum from a fish paratenic host and in the cystacanth stage of Echinorhynchus gadi from an amphipod intermediate host was investigated by means of transmission electron microscopy. The species were obtained from hosts living in the Baltic Sea. The observations indicate that ovarian development was more advanced in cystacanths of E. gadi than in juveniles of C. semerme and C. strumosum. Free ovaries were present in E. gadi with an obvious supporting syncytium and surface microvilli. The biological significance of the different stages of ovarian development in these species is discussed briefly in relation to either reproductive strategies and host specificity in definitive hosts or to the attainment of infectivity by cystacanths in intermediate hosts.

Genetic proximity between two karyotypic groups of Trypanosoma congolense was investigated using as hybridization probes: (i) total genomic DNA, (ii) a 35 nucleotide long synthetic oligonucleotide, and (iii) non-variant antigen type (non-VAT) specific complementary DNAs. The phylogenetic relationship between Trypanosoma brucei and T. evansi, both of which are accepted species in the subgenus Trypanozoon, was used as a reference to assess the phylogenetic proximity of the two groups of T. congolense. Results indicate that some morphologically indistinguishable T. congolense populations differ in a variety of molecular and genetic properties: molecular karyotypes, majority of the DNA sequences, and the restriction enzyme sites in the genomic environments of various conserved genes. The implications of these findings for trypanosome evolution and T. congolense epidemiology are discussed.

The total number and distribution of schistosomula of Schistosoma mansoni and S. haematobium in all tissues and organs of mice from infection to 14–27 days was determined by compressed tissue autoradiography. Attrition of schistosomula, manifested as a decrease in the number of autoradiographic foci, was observed in organs other than the liver. Attrition commenced about 2 days after cercariae entered the skin, and conformed to a single exponential function with a rate constant (± standard error) of 7·0±0·5%/day for S. mansoni and 3·2±0·7%/day for S. haematobium. The temporal distribution of schistosomula of S. mansoni and S. haematobium differed quantitatively. In the case of S. mansoni, concomittant with a decrease in skin counts, the lung curve rose rapidly to a peak centred on day 6 and thereafter decreased more or less parallel to the total body curve. Significant accumulation in the liver was not observed until day 7, whereupon liver counts rose steadily to a plateau that extended from about day 14 to the end of the experiment and approximated the number of adult worms recovered from the hepatic portal vessels on day 42. A maximum of 26% and mean of 12% of all foci in the body were counted on autoradiograms of tissues other than the skin, lung and liver. The pelt averaged 14% of the body weight yet schistosomula were detected only in the area initially exposed to cercariae. The eviscerated carcass averaged 54% of the body weight yet contained only 0·8% −3·4 % of the schistosomula during the period of accumulation in the liver. Between day 6 and day 14, the ratio of schistosomula in the pulmonary circulation to schistosomula in the systemic circulation did not remain constant, as would be the case if schistosomula circulated passively and randomly, but instead displayed a statistically significant decrease from 0·92 and 0·85. For these reasons, it was considered unlikely that schistosomula had circulated randomly and repeatedly through the pulmonary and systemic circulations and entered the hepatic portal system by chance, as hypothesized by Miller & Wilson (1980). Instead it was considered more probable that schistosomula migrating from lungs to liver had followed a directed path through intervening vessels (Kruger, Heitman, van Wyk & McCully, 1969) or tissues (Wilks, 1967). Schistosoma haematobium distribution patterns differed from those of S. mansoni in slower movement of schistosomula from skin, their peak accumulation in lungs at about day 8 followed by a low rate of decrease, and a very low liver plateau which approximated the number of adult worms recovered from the hepatic portal vessels on day 42.

The major transaminase in Heligmosomoides polygyrus, Panagrellus redivivus and rat liver was the 2-oxoglutarate-glutamate system, with relatively few amino acids acting as donors for the pyruvate-alanine and oxaloacetate–aspartate systems. The relative effectiveness of the different amino acid donors in the three transaminase systems was similar in all three tissues. Both H. polygyrus and P. redivivus can oxidatively deaminate a range of L-amino acids, although D-amino acid oxidase activity was low. Serine and threonine dehydratase activity and histidase activity were present in H. polygyrus and P. redivivus and both nematodes were also able to deaminate glutamine, asparagine and arginine. When NAD(H) was the cofactor the glutamate dehydrogenases of H. polygyrus and P. redivivus showed similar regulatory properties to the mammalian enzyme. However, with NADP(H) the results were anomalous. The capacity of both nematodes to transaminate and oxidatively deaminate amino acids was broadly similar and comparable to mammalian tissue. Glutamate dehydrogenase is probably the major route for deamination in these nematodes. A complete sequence of urea cycle enzymes could not be demonstrated in either P. redivivus or H. polygyrus.

The relationship between infection rates of rainbow trout (Salmo gairdneri) by Diplostomum spathaceum and cercarial concentration, water flow rate and temperature were investigated by means of controlled infections within a flume. A linear relationship was obtained between cercarial concentration and mean abundance of metacercariae/fish. A biphasic relationship occurred between flow rate and abundance of metacercariae. Within the confines of the flume, it was possible to control the infection rate of trout with D. spathaceum cercariae by manipulating flow rate, suggesting that it may be a possible method of controlling diplostomiasis on fish farms. No infection occurred in fish infected and maintained below 10°C and the optimum infection temperature was approximately 17·5°C. Infections became established in fish infected at 7·5 and 5°C but maintained at 15°C prior to examination. Trout were infected at 7·5°C for 10–50 min and all attached cercariae were washed off and removed from the flume. Following infection, fish were either maintained at 7·5°C or 15°C prior to examination. Using this method, it was possible to ascertain that it was migration and not penetration which was inhibited at water temperatures below 10°C.

All of the enzymes of proline catabolism were present in Heligmosomoides polygyrus and Panagrellus redivivus and the activities were, in general, similar to those found in rat liver. Both nematodes were also shown to be able to catabolize the branched-chain amino acids leucine, isoleucine and valine, by pathways similar to those found in mammalian liver. There were no significant differences in amino acid catabolism between the animal-parasitic and free-living species of nematode.
