SRY is a unique gene on the Y chromosome in most mammalian species including the laboratory mouse, Mus musculus, and the closely related European wild mouse species M. spicilegus, M. macedonicus, and M. spretus. In contrast, SRY is present in 2–6 copies in the more distantly related Asian mouse species M. caroli, M. cervicolor, and M. cookii and in 2–13 copies in the related murid species Pyromys saxicola, Coelomys pahari, Nannomys minutoides, Mastomys natalensis, and Rattus norvegicus. Copy numbers do not correlate with known phylogenetic relationships suggesting that SRY has undergone a rapid and complex evolution in these species. SRY was recently proposed as a molecular probe for phylogenetic inferences. The presence of multiple SRY genes in a wide range of murid species and genera, and at least one cricetid species, necessitates caution in the use of SRY for phylogenetic studies in the Rodentia unless it is ascertained that multiple SRY genes do not exist.

A population genetic model is developed and then applied to the synonymous gene sequence variation observed in samples of the Human Immunodeficiency Virus Type 1 (HIV-1). The samples, which were taken from several previous studies, contain sequences of the envelope glycoprotein gene (gp 120) of HIV-1. This analysis suggests that the viral population within an infected patient at any specific time is likely to be composed of close relatives. The viruses in a sample are likely to share a recent common ancestor probably due to consistent positive selection for non-synonymous mutations coupled with low recombination in this region of the genome. There is no substantial difference in synonymous evolutionary rate between samples of sequences obtained from Peripheral Blood Mononucleate Cells (PBMCs) and samples taken from blood plasma. This is likely to be due to the high rate of migration between these 2 HIV sub-populations. The mutation rate for the genetic region examined is estimated at 9·20 × 10−4 per site per month. Under the assumptions of the estimation procedure, this estimate can be bounded between 8·50 and 9·91 × 10−4 with 95% confidence. When coupled with direct estimates of mutation rate, the rate of synonymous evolution suggests that the mean number of generations per month for HIV-1 in vivo is between 1 and 4.

We consider genetic models for the inheritance of the particular colour patterns of silver foxes. The models are evaluated by computation of statistical likelihoods based on observations of related foxes in extended pedigrees. Problems caused by incomplete paternity information are addressed by inferences based on phenotypic observations. The unreliability of subjective evaluations of fur colour also provides difficulty, in particular crossfoxes emerge as being difficult to differentiate. No evidence of linkage between Agouti locus and Extension locus is found in this dataset.

Chiasma distributions along bivalents 1 and 14 in female and male mice were studied. It was shown that the average chiasma number in both chromosomes show no sex difference. There are however, significant sex differences in chiasma distribution along 1 and 14 chromosomes. In males there are two terminal chiasma peaks in chromosome 1 and one subtelomeric peak of chiasmata in chromosome 14. In females chiasma distributions are more even. According to genetic data, females produce more recombinants between loci of chromosome 1 than males do. By means of a computer simulation it was demonstrated that the differences in the average recombination frequency result from differences in chiasma distribution.

The genome of the house fly, Musca domestica, contains an active transposable element system, called Hermes. Using PCR and inverse PCR we amplified and sequenced overlapping segments of several Hermes elements and from these data we have constructed a 2749 bp consensus Hermes DNA sequence. Hermes termini are composed of 17 bp imperfect inverted repeats that are almost identical to the inverted terminal repeats of the hobo element of Drosophila melanogaster. Full length Hermes elements contain a single long ORF capable of encoding a protein of 612 amino acids which is 55% identical to the amino acid sequence of the hobo transposase. Comparison of the ends of the Hermes and hobo elements to those of the Ac element of Zea mays, and the Tam3 element of Antirrhinum majus, as well as several other plant and insect elements, revealed a conserved terminal sequence motif. Thus Hermes is clearly a member of the hobo, Ac and Tam3 (hAT) transposable element family, other members of which include the Tagl element from Arabidopsis thaliana and the Bg element from Zea mays. The evolution of this class of transposable elements and the potential utility of Hermes as a genetic tool in M. domestica and related species are discussed.

Susceptibility to urethane-induced lung adenomas in mice has a polygenic mode of inheritance, with no obvious discontinuity in lung tumour counts among 37 AXB recombinant inbred strains. However, mean tumour counts were markedly higher in strains carrying the A/J allele at the Kras2 and H2 complex than in those carrying the C57BL/6 allele. In 162 F2 hybrids and small numbers of both backcrosses between strain A/J (susceptible) and C57BL/6 (resistant) mice, five factors influencing susceptibility were identified. Variation due to the ‘major’ Kras2 locus (chromosome 6) accounted for 60% of the total variation. ‘Minor’ loci linked to microsatellite markers Tnfb (in the H2 complex), D9Mit11 and D19MH16 (on chromosomes 17, 9 and 19, respectively) accounted for a further 13% of the variation, and males had more tumours than females with sex differences accounting for 2% of the variation. No significant association with 32 other loci was detected. On a square-root transformed scale, heterozygotes at all marker loci were of intermediate susceptibility compared with homozygotes. Thethree minor loci and sex only affected lung tumour counts when at least one susceptible Kras2 allele was present.

In order to understand the evolution of female mate preferences it is important to determine whether the genes for the preference and those for the preferred character are linked. It has previously been shown that female preference in the seaweed fly, Coelopa frigida, varies with the αβ inversion system on chromosome I. This inversion system is known to genetically determine, at least in part, the male preferred character, large size. This study was undertaken to determine whether the genes determining mate preferences, as well as those determining female receptivity, co-inherit with the inversion. In the full sibs of animals recently collected from a natural population in Sweden it is shown that high acceptance rate and strong preference for large male size both co-segregate with the α form of the inversion, and that low acceptance rate and a weak preference for large size co-segregate with the β form of the inversion. Both sets of genes appear to be located in or near the αβ inversion. The heterogeneity between crosses suggests the natural population from which the animals were collected was polymorphic for behavioural genes on the β haplotype. Crosses involving animals that had been in laboratory culture for seven generations indicated that variation in female mating behaviour had been lost. Possible reasons for the apparent instability of such behaviour are discussed.

Over twenty distinct families of long terminal direct repeat (LTR)-containing retrotransposons have been identified in Drosophila melanogaster. While there have been extensive analyses of retrotransposon transcription in cultured cells, there have been few studies of the spatial expression of retrotransposons during normal development. Here we report a detailed analysis of the spatial expression patterns of fifteen families of retrotransposons during Drosophila melanogaster embryogenesis (17.6, 297, 412, 1731, 3S18, blood, copia, gypsy, HMS Beagle, Kermit/flea, mdg1, mdg3, opus, roo/B104 and springer). In each case, analyses were carried out in from two to four wild-type strains. Since the chromosomal insertion sites of any particular family of retrotransposons vary widely among wild-type strains, a spatial expression pattern that is conserved among strains is likely to have been generated through interaction of host transcription factors with cis-regulatory elements resident in the retrotransposons themselves. All fifteen families of retrotransposons showed conserved patterns of spatially and temporally regulated expression during embryogenesis. These results suggest that all families of retrotransposons carry cis-acting elements that control their spatial and temporal expression patterns. Thus, transposition of a retrotransposon into or near a particular host gene-possibly followed by an excision event leaving behind the retrotransposon's cis-regulatory sequences-might impose novel developmental control on such a host gene. Such a mechanism would serve to confer evolutionarily significant alterations in the spatio-temporal control of gene expression.

The total genomic copy numbers of ten families of transposable elements of Drosophila melanogaster in a set of ten isogenic lines derived from a natural population were estimated by slot-blotting. The numbers of euchromatic copies of members of each family were determined for each line by in situ hybridization of element probes to polytene chromosomes. Heterochromatic numbers were estimated by subtraction of the euchromatic counts from the total numbers. There was considerable variation between element families and lines in heterochromatic abundances, and the variance between lines for many elements was much greater for the heterochromatin than for the euchromatin. The data are consistent with the view that much of the β-heterochromatin consists of sequences derived from transposable elements. They are also consistent with the hypothesis that similar evolutionary forces control element abundances in both the euchromatin and heterochromatin, although amplification of inert sequences derived from transposable elements may be in part responsible for their accumulation in heterochromatin.

A phenotypic analysis was performed on two alleles at the pygmy locus which arose by insertional mutagenesis in transgenic mice. Similar to the spontaneous mutant pygmy, the adult insertional transgenic mutants are 40% of the size of wild-type litter-mates whereas adult heterozygotes are 80% of wild-type litter-mates. An analysis of the various organs revealed that, in general, there was a reduction in weight of each organ commensurate with the overall reduction in body size. However, two organs did not follow this pattern, the brain being disproportionately larger and the adrenals disproportionately smaller in the mutant mice. In addition, mini-mice have less adipose tissue than their wild-type or heterozygous litter-mates. A developmental analysis determined that mutants could first be identified on the basis of reduced body weight at day 15·5 of gestation. The small size is not due to a growth hormone deficiency so these mice differ from other known dwarf mouse mutants. Therefore they should provide insight into the growth hormone-resistant human dwarnsms and help in furthering our knowledge of mammalian growth and development.

A total of 357 house mice (Mus domesticus) from 83 localities uniformly distributed throughout Switzerland were screened for the presence of a homogenously staining region (HSR) on chromosome 1. Altogether 47 mice from 11 localities were HSR/ + or HSR/HSR. One sample of 11 individuals all had an HSR/HSR karyotype. Almost all mice with the variant were collected from the Rhone valley (HSR frequency: 61%) and Val Bregaglia (HSR frequency: 81%). For samples from most of thearea of Switzerland, the HSR was absent. There was no strong association between the geographic distribution of the HSR and the areas of occurrence of metacentrics. However, at Chiggiogna the HSR was found on Rb (1·3). Possible explanations for the HSR polymorphism are discussed.

A hypothesis has been tested that alterations in the molecule of histone H1 are capable of influencing quantitative traits of an organism. Two pairs of isogenic lines were constructed by selfing of plants kept heterozygous either for allelic variants of histone H1 subtype 1 (His1 gene) or for allelic combinations (haplotypes) of closely linked genes of H1 subtypes 3, 4, 5 and 6 (gene cluster His(2–6)). After 19 and 15 generations of selfing, respectively, expectation of the length of chromosome which remained heterozygous has comprised 2·6 cM near Hisl and 3·5 cM near His(2–6). The third pair of isogenic lines was obtained as a result of two successive intracluster cross-over events bordering the gene of the subtype 5 (His5). In each pair of isogenic lines there have been revealed some statistically significant differences between mean values of a number of quantitative traits.

Two approaches to the evolution of phenotypic plasticity in heterogeneous environments have recently been put forward. The first focuses on selection on the character expression within each environment; plasticity is seen as a by-product of local selection in various habitats. The second approach focuses on selection on the parameters of the response function of genotypes, and selection is thought to change the frequencies of ‘plasticity’ genes that affect the function. This paper discusses the relationship between the two approaches, with emphasis on applications. A method is described that allows switching from one approach to the other. It is argued that character state and reaction norm approaches, while to a large extent interchangeable, usually differ in the response function chosen. This choice, however, may strongly affect the biological interpretation. The methods outlined in this paper permit one to look at the data from different perspectives in order to avoid this danger.

The probability of fixation of a mutation with selective advantage s will be reduced by substitutions at other loci. The effect of a single substitution, with selective advantage , can be approximated as a sudden reduction in the frequency of the favourable allele, by a fraction w = 1 −(s/S)r/s (where r is the recombination rate). An expression for the effect of a given sequence of such catastrophes is derived. This also applies to the ecological prxoblem of finding the probability that a small population will survive, despite occasional disasters. It is shown that if substitutions occur at a rate Δ, and are scattered randomly over a genetic map of length R, then an allele is unlikely to be fixed if its advantage is less than a critical value, Scrit = (π2/6)(2ΔS/(Rlog(S/s))). This threshold depends primarily on the variance in fitness per unit map length dueto substitutions, var(W)/R = 2ΔS/R. With no recombination, the fixation probability can be calculated for a finite population. If Δ > s, it is of the same order as for a neutral allele ( ≈ Δ/(2N(Δ−s))), whilst if , fixation probability is much higher than for a neutral allele, but much lower than in the absence of hitch-hiking . These results suggest that hitch-hiking may substantially impede the accumulation of weakly favoured adaptations.

Up to now, no case of spontaneous sex reversal has been described in the urodele amphibian Pleurodeles waltl reared at ambient temperature. However in offspring reared under laboratory conditions, males, females but also intersex individuals were obtained. The males, some females and the intersexes had the ZZ male genotype identified through a test performed with a sex-linked enzyme. The other females had the ZW female genotype. Using the animals of this particular strain, an offspring analysis was made on crosses between, respectively, ZZ males and ZW females, ZZ males and ZZ females, and ZZ intersexes and ZZ males or females. All these crosses gave ZZ males, ZZ females and ZZ intersexes. The spontaneous complete or partial sex reversal is inheritable, but the genetic mechanism of this inheritance has not yet been elucidated.

The substructural organization of chromosome cores or nonhistone proteins was studied within intact metaphase chromosomes at the second meiotic division in the grasshopper Trilophidia annulata by silver staining as well as light microscopy and whole mount electron microscopy of squash chromosomes. Our results revealed that the metaphase II chromosome contains a longitudinal, helical coiling core structure. Probably the two last organizational levels of the core packaging are achieved by helical coiling. The core structure retains the morphological characteristics of the original metaphase chromosome, surrounded by a halo of dispersed materials, which may be composed mainly of nonhistone proteins. The kinetochore is found to be connected with the chromosome core. The present findings combined with our previous observations on the helical structure of metaphase II chromosomes suggest that the folding path of the internal core structure in metaphase chromosomes is consistent with the final helical arrangement of the chromosome itself. These observations also imply that in condensed metaphase chromosomes nonhistone protein may form a compact network structure with helical appearance, which extends throughout the entire chromosome.

Marker Assisted Selection (MAS) based on additive effects associated with alleles at marker loci, estimated by linear regression of individual phenotype on the markers, was applied to characters with non-additive gene action and non-additive environment. The base population was the F2 generation of a cross between two inbred lines. Computer simulations of MAS were conducted for characters with dominance, epistasis and genotype-environment interaction approximated by the ‘additive-multiplicative’ model. MAS was more effective than purely phenotypic selection in all cases. The efficiency of MAS for characters with non-additive gene actionis comparable to (and for negative dominance even higher than) the efficiency of MAS for strictly additive characters. Environmental non-additivity, however, lowers the efficiency of MAS. Almost all results concerning the efficiency of MAS in our previoussimulations of purelyadditive traits are applicable to non-additive traits.

We develop a model to predict the increase in genetic variance of a quantitative character in a hybrid population produced by crossing two previously isolated populations of the same species. The increase in variance in the F2 hybrids, the ‘segregation variance’, is caused by differences in the average allelic effects at each locus and by linkage disequilibrium among loci. We focus on the case in which the character is additively based and the average value of the character does not differ in the two populations. In that case the predicted segregation variance depends strongly on what is assumed about the genetic basis of the character. If the genetic variance of the character in each population is attributable to loci with numerous alleles of small effect that are in moderate frequency, as in Lande's (1975) model, the segregation variance should increase linearly with time since the populations were isolated, at a rate determined by the inverse of the effective population size. If the genetic variance is attributable to loci with alleles in very low frequency, as in Turelli's (1984) house-of-cards model or in Barton's (1990) model of pleiotropic, deleterious alleles, then the segregation variance in the hybrid population increases at a much lower rate.

Time-dependent behaviour of linkage disequilibrium when there was initial linkage disequilibrium is studied in a finite island model assuming neutrality. Explicit expressions for linkage disequilibrium parameters are obtained. From these expressions, the initial and the ultimate decay rates of linkage disequilibrium parameters are found to be increased and decreased, respectively, by finiteness of the population when recombination rate, migration rate and inverse of subpopulation size are of comparable order. Thus, linkage disequilibrium created in the past may persist longerin smaller subdivided populations. Also, differentiation of the gametic parameter of linkage disequilibrium among subpopulations is found to diminish quickly compared tothe linkage disequilibrium in the whole population. Implications of these results for the interpretation of linkage disequilibria in natural populations are discussed.
