Professor Gwendolen Rees, or Gwen as she prefers to be known, was educated at the Intermediate School for Girls, Aberdare, between 1918 and 1924. Aberdare, a valley community in South Wales was, in the nineteenth century, world-famous as a centre for the Welsh iron and coal industry. Unlike its neighbouring town, Merthyr, however, it was not totally dominated by industry and it is especially renowned for its spacious and beautiful park near the town centre. During Gwen's early life, Aberdare was one of the largest and most influential centres of literary culture in Wales, and a place where education was of prime importance in the community. As far back as 1867, Aberdare men and women had claimed that ‘what we think today, Wales will think tomorrow’. Could these be the reasons why Aberdare has produced so many distinguished academics? None, however, has achieved greater distinction than Gwendolen Rees who, in 1971, became the first woman academic in Wales to be elected a Fellow of the Royal Society. Since she has often been affectionately referred to as ‘one in a million’ by her former students, the metaphor assumed literal significance in 1971, when the female population of Wales was 1446000. Because of a phenomenal life-long dedication to her chosen career as university teacher, researcher and administrator, together with a genuine lack of desire for personal acclaim, there may have been many people who had never heard of her until 1971.

Oocysts of fowl coccidia, suspended in a 12% aqueous solution of alginic acid, were dropped into 5% calcium chloride solution to form beads. Following ingestion by chicks, excystation of sporozoites was not affected. Oocyst viability was best when beads were air-dried to 33% of their original weight and stored in airtight containers at 4°C. Infectivity was fully maintained for 8 weeks. Trickle infection, using beads administered orally to 2- or 3-week-old chickens, or mixed daily in food of chicks from 1-day-old, established a high level of immunity. This method of oocyst administration offers some practical advantages for the conduct of immunity and chemotherapy experiments.

We have compared the surface radio-iodinated proteins of uninfected and Plasmodium falciparum-infected erythrocytes from natural infections of human patients. Cryopreserved infected blood from Gambian children with falciparum malaria was thawed, cultured to the middle trophozoite stage, and surface radio-iodinated. Trophozoite-infected cells were enriched about 10-fold on a Percoll gradient newly designed to separate cells based on their differential permeability to sorbitol. Infected blood was radio-iodinated and erythrocyes from the fraction enriched in parasitized cells and uninfected erythrocytes from the same sample obtained from the gradient and compared by SDS–PAGE and autoradiography. In each sample, parasitized erythrocytes contained one or more polypeptides of very high molecular weight (Mr 250000–300000) that were not found on uninfected erythrocytes from the same patient. These proteins were isolate-specific in size and number, suggesting that natural isolates contain a variable number of different P. falciparum phenotypes for this surface protein. In addition, these radio-iodinated surface proteins could not be extracted from the host cell membrane by the non-ionic detergent Triton X-100, but were extracted by SDS. The properties of these proteins suggest they are the equivalent for natural infections of the strain-dependent antigen previously described (Leech, Barnwell, Miller & Howard, 1984) on the surface of P. falciparum-infected Aotus erythrocytes. In addition, we observed a second parasite-dependent modification of labelled proteins on infected erythrocytes with the appearance of a new band of Mr 30000. There were also variations in the pattern of radio-isotope labelled proteins on uninfected erythrocytes from different patients.

The effects of malaria infection on RBC sialic acids and sialoglycoproteins were studied with asexual blood-stage infections of Plasmodium knowlesi in rhesus monkeys. Glycoprotein radio-isotope labelling methods were used to compare the sialoglycoproteins of normal RBC and P. knowlesi schizont-infected RBC (SI-RBC). Tritiation of glycoproteins from SI-RBC with the standard sialidase + galactose oxidase/NaB3H4 method or standard periodate/NaB3H4 method was significantly decreased when compared to normal RBC. However, tritium uptake into glycoproteins was normal when SI-RBC were treated with 5-fold higher concentrations of both enzymes in the first labelling method, or with a 5-fold increase in the molar ratio of periodate to sialic acid in the second method. The mobility of tritiated host cell glycoproteins on SDS–polyacrylamide gels was identical for SI-RBC and normal RBC. New bands, possibly glycoproteins, of 230, 160, 90, 52, and 30 kDa were detected after labelling SI-RBC by the modified periodate/NaB3H4 method. Sialic acid analysis of normal rhesus monkey RBC (62μg/1010 RBC) revealed that 46% of the total sialic acid was N-glycolylneuraminic acid, 33% was N-acetyl-9-O-acetylneuraminic acid, and the remainder N-acetylneuraminic acid. SI-RBC collected either directly from infected monkeys or after in vitro culture of ring-infected RBC in horse serum, had increased total sialic acid (126 or 115μg/1010 RBC, respectively). The sialic acid content of infected RBC must increase during parasite development since RBC infected with ring-stage P. knowlesi had the same content as normal RBC. There was no significant difference in the ratio of the three sialic acids of SI-RBC and normal RBC. In contrast, the uninfected RBC from infected blood of different monkeys showed marked variation in sialic acid composition and generally had a lower sialic acid content than normal RBC.

Schizont-infected red blood cells (SI-RBC) from Plasmodium berghei-infected mice contain between 2 and 10 times as much sialic acid as uninfected RBC from the same blood (99–550 μg/1010 RBC versus 33–65 μg/1010 RBC). Total RBC samples from infected animals containing up to 63% ring- and trophozoite-infected cells had identical sialic acid contents to purified RBC samples (of < 3% parasitaemia) from the same blood (52–64 μg/1010 RBC). We conclude that RBC containing immature parasites have the same sialic acid content as uninfected RBC from infected blood and that total cellular sialic acid increases during maturation to the schizont stage. Uninfected RBC from infected blood had 25–50% as much sialic acid as normal mouse RBC (33–65 μg/1010 RBC versus 126 μg/1010 RBC). There were no qualitative changes in RBC sialic acids, all RBC samples having 60–70% N-acetyl neuraminic acid, 30–40% N-acetyl-9-O-acetylneuraminic acid and 5–10% N-gly colylneuraminic acid. The quantitative changes we observed during infection must reflect changes in murine sialoglycoconjugates, as we have shown elsewhere that Plasmodia do not synthesize or contain sialic acids. Since the sialic acid composition of mouse serum glycoconjugates is quite different to that of the RBC fractions studied here, the quantitative data suggest that part of the sialic acids of the uninfected RBC has been transferred to SI-RBC. With higher molar ratios of periodate to substrate than generally used, we were able to radio-isotopically label normal murine sialoglycoproteins on SI-RBC and purified uninfected RBC from infected blood by the periodate/NaB3H4 method. Several new proteins were then tritiated with SI-RBC but these proteins may be intracellular and could even lack sialie acid.

A study has been made of the damage incurred by normal and Plasmodium falciparum-infected human erythrocytes following exposure to a variety of oxidant-generating systems. Hydrogen peroxide, produced by the glucose–glucose oxidase system, increased methaemoglobin formation within normal erythrocytes while normal levels of oxyhaemoglobin were maintained. Exposure to products of the xanthine–xanthine oxidase interaction did not have the same effect. Malondialdehyde measurements indicated that the host cell membranes of parasitized cells had undergone lipid peroxidation even before exposure to the oxidant-generating systems. Lipid peroxidation of normal and parasitized cell membranes was increased upon exposure to reagent-grade hydrogen peroxide and alloxan: this increase was not observed following exposure to the two enzyme–substrate systems that generated reactive oxygen intermediates. In addition, the effects of parasitism on intracellular levels of catalase and superoxide dismutase were assessed. Normal and parasitized erythrocytes were found to possess similar levels of these enzymes, which protect against oxidant-induced damage. It was therefore concluded that the increased susceptibility of infected cells to oxidant damage was probably not related to any decrease in the function of these enzymes.

Sera from 40 onchocerciasis patients from the Yemen Arab Republic with either mild localized forms of onchocerciasis, intermediate or severe localized forms of the disease or generalized forms of infection were studies with rspect to their IgG and IgM response against Onchocerca volvulus antigens. Immunoblotting, performed with SDS–PAGE-separated proteins of female O. volvulus and quantified by densitometric scanning, revealed IgG and IgM antibodies against worm components in sera of all patients. Persons with intermediate or severe localized forms of onchocerciasis had a stronger IgG response against more proteins than individuals of the other group. However, some antigens (Mr 21, 23, 30, 33 kDa) induced comparable quantities of IgG in all groups. The IgM response of patients with mild localized forms of onchocerciasis was more intensive and directed against more antigensthan in the other groups. No antigens were detected that were recognized only by individuals with low levels of microfilaridermia. In all groups, varying concentrations of antibodies against cuticle, muscle/hypodermis layer and/or uterus of female O. volvulus were detected by the indirect immunofluorescence test using frozen worm sections as antigen. The highest mean antibody titres were found in patients with intermediate and severe localized forms of disease.

Antigenic variation in the ILDar 1 serodeme of the naturally rodent-infective stock of West African Trypanosoma vivax has been investigated following cyclical transmission. The immunofluorescent and immune lysis tests were employed with a panel of 39 variant-specific mouse antisera. When antigenically homogeneous, or mixed, populations were transmitted by tsetse flies to goats, the first peak parasitaemias arising in the goats were antigenic mixtures (up to 9 major, and several minor variants being recognized in some cases) from which the ingested variant was absent. Although first peak parasitaemias in similarly infected goats showed some variants in common, there was no obvious relationship between the VAT profiles in different goats. When these populations were expanded in irradiated mice, VAT heterogeneity was maintained with a tendency towards the development of predominant variants in some, but not all, instances. Six additional variants, derived following the growth of bloodstream form ILDat 1·9 in 37°C culture, were also represented in goat and mouse populations. Two further variants, isolated after cyclical development of ILDat 1·9-derived trypanosomes in vitro, were not present in the early parasitaemias in goats and mice.

The transmission of infection with Giardia muris among CD-I mice was quantified by determining the duration of the latent period and intensity of cyst release during the acute phase of the infection. Housing of experimentally inoculated mice with uninfected mice in the same cage resulted in 100% transmission of infection. The dissemination of the infection was faster if experimentally inoculated hosts were passing high numbers of cysts at the time of housing with uninfected animals. The course of ‘naturally’ acquired and experimentally induced infections was similar. We conclude that G. muris has a high transmission potential and that the number of cysts randomly consumed does not influence the course and eventual outcome of the infection.

The numerical distributions of Echinococcus granulosus in an experimental dog population are described. At all dose rates of protoscoleces from 10 to 175000 the distribution of worms was over-dispersed. Host age had no effect. There was a direct proportionality between the infective-stage density and rate of infection, and between the latter and the index of clumping. The worm burdens were significantly higher in the proximal than distal portions of the small intestine. Lengths of the 3- and 4-segmented worms increased from 4 to 10 and 4 to 8 weeks of age, respectively. Thereafter apolysis was asynchronous and could not be determined. Eggs were first detected in the faeces at 6 weeks and the mean age at oogenesis was 7·26 weeks. Retarded growth of the whole population of worms was observed in some dogs. For the first few infections, worm burdens varied widely in the same dog, but by the 6th infection 50% of the dog population had developed a relative insusceptibility to infection. Growth or oogenesis of the worms were not affected. A short-acting immune response was artificially induced in some dogs following the parenteral injection of activated embryos of E. granulosus, Taenia hydatigena, T. ovis, T. multiceps, T. pisiformis and T. serialis. The response affected either the number of worms established, growth or oogenesis or all three parameters. There was a strong positive correlation between numbers and lengths of worms in dogs with acquired and induced immunity, indicating that no ‘crowding’ effects were involved. In sheep populations the mean number of cysts which established was directly proportional to the number of eggs given, implying that there was no negative feedback mechanism operating at this stage of the life-cycle. The distribution of the larval population in sheep was over-dispersed and the index of clumping increased with the size of the egg dose from 25 to 2500 eggs. Protoscoleces were first observed in cysts at 2 years and the proportion producing them increased with age, with an estimate of 50% of cysts containing protoscoleces at 6 years. No deaths were observed in dogs or sheep even when high parasite burdens were present, implying that E. granulosus does not regulate the population of its hosts.

A mathematical model of the life-cycle of Echinococcus granulosus in dogs and sheep in New Zealand is constructed and used to discuss previously published experimental and survey data. The model is then used to describe the dynamics of transmission of the parasite, and the means by which it may be destabilized. It is found that under the conditions that prevailed in New Zealand during the late 1950s, at the time of surveys of this parasite, the dog–sheep life-cycle was not regulated by any effective density-dependent constraint. In contrast there was evidence for an effective acquisition of immunity to reinfection by cattle. The long time to maturity of the cyst in sheep, together with the practice of feeding aged sheep to dogs, provides a time delay in the intermediate host. By comparison, the time to maturity of the adult stage in dogs is short, but it is of sufficient magnitude to be a key factor in the destabilization of the system by a regular dog-dosing programme. The model used to describe the life-cycle is a linear integrodifferential equation of the Volterra type. Such equations are intrinsically unstable in that a small perturbation in parameters can drive a previous equilibrium solution to zero. At the time of the surveys, the value of the basic reproductive rate, R0 was close to 1, and it has since been reduced below 1 by control measures.

We present an analysis of the survival and migration rates of the infective (L3) stages of Oslertagia ostertagi and Cooperia oncophora. Although the majority of laboratory studies show that the survival of the L3 stage depends upon temperature, moisture and the age of the larvae, a simple mathematical model of larval demography, in which their mortality and migration rates are held constant, provides as good agreement between observed and predicted larval counts as models in which these rate processes are made explicit functions of larval age and microclimate. Maximum-likelihood estimates of larval mortality rates in the faeces and on the herbage are 0·0284/day and 0·00887/day respectively. The average migration rate from faeces to herbage under temperate Northern European conditions is estimated as 0·00884/day. Finally, we discuss the probable scale of L3 larval losses due to desiccation and lavage (active or passsive migration into the soil).

The tegumental surface of Schistosoma mansoni sporocysts is the site of both nutritive and immunological interactions with haemolymph cells and plasma of Biomphalaria glabrata, the schistosome intermediate host. Within minutes of being placed in host plasma, sporocysts acquire plasma antigens, and within 3 h host plasma antigens are present on the surface at near steady state. Though a wide variety of peptides is acquired, there is selection. Furthermore, some differences occur in the peptides acquired from the plasma of susceptible and resistant strains of snail. Acquired antigens are rapidly processed, and are predominantly undetectable in tegumental extracts after a few hours. In contrast, rabbit antibody on sporocysts remains in situ for at least 48 h, so under some conditions there is stable expression of certain tegumental antigenic determinants.These data, obtained using antibodies to snail plasma antigens and to sporocyst tegumental antigens, are discussed in the light of current ideas on the cellular and molecular basis of susceptibility and resistance in this host#parasite system.

Interactions between tapeworm species in a single host offer intriguing opportunities for immunological studies that attempt to identify the mechanism(s) underlying protection against cestode infections. Mice that are immunized against Hymenolepis citelli infections were shown to be refractory to subsequent H. diminuta challenge infections. The reciprocity of the response was also demonstrated, although the protection recorded for H. diminuta when mice are sensitized with H. citelli is weaker than that observed when mice are primed with H. diminuta against H. citelli challenge. H. citelli was also shown to be expelled simultaneously during the rejection phase of H. diminuta in concurrent infections, indicating the susceptibility of the former tapeworm to the rejection mechanism initiated by the latter. H. microstoma immunized mice were shown to be strongly protected against heterologous H. citelli challenge. However, mice primed against H. citelli were not as strongly protected against H. microstoma challenge infections: a statistically significant protection was obtained only after a 12-cysticercoid H. citelli primary infection, although a 6-cyst infection did stunt the growth of H. microstoma challenge worms. It is presently suggested that the cross-protective responses observed in the study between H. citelli, H. diminuta and H. microstoma may have emanated from a specific immunological cross-reactivity due to the sharing of similar immunogens.

Two simple methods of measuring the levels of inequality in reproductive success of different individuals in parasite populations are presented. These techniques are then applied to a number of sets of data for cestodes and acanthocephalans. The analysis suggests that both population density and host nutrition are important in determining the observed degree of inequality in reproductive success and body size. Cestodes, with a more flexible growth form, are shown to exhibit higher levels of inequality than acanthocephalans. The discussion outlines the evolutionary importance of considering variation in the reproductive success of different individuals.

Using a range of infection techniques, the ability of cercariae of the ectoparasitic digenean Transversotrema patialense to infect 24 species of tropical freshwater fish is examined. Of the 24 species (from 11 families) it is possible to infect 21 (from 8 families) under laboratory conditions. Reproductive success (R) for T. patialense from the cercarial infection process to the production of viable eggs by adult flukes is estimated for infections on two fish hosts, the cyprinid Brwhydanio rerio and the characinid Cheirodon sp. in controlled laboratory conditions. To calculate R, separate estimations of parasite infectivity, survival, fecundity and egg viability were made on populations of both host species. The enhanced reproductive success (R) of T. patialense on Cheirodon sp. is the composite resultant of higher infectivity, lower mean expected adult fluke life-span and higher mean fecundity/surviving worm when compared with infections on B. rerio under identical experimental conditions. Different patterns of egg viability of the two hosts do not materially influence differential reproductive success. A variety of formal techniques for handling the interacting components of reproductive success are described. The relationship between these techniques and the concept of basic reproductive rate, R0 (Macdonald, 1957; Anderson, 1982) is assessed.

An epidemiological survey of the distribution and abundance of Diplostomum spathaceum in farmed rainbow trout (Salmo gairdneri) from Mill of Cantray trout farm (Nairn, Morayshire) involved monthly sampling of fish over a 3-year period. Diplostomurn metacercariae were present in both the lens and aqueous humour of infected fish, and these have been treated separately throughout the study. The infection period was normally between May and September each year and transmission of the parasite from snail to fish did not occur at temperatures below 10°C. The prevalence and abundance of both lens and humour metacercariae reached a maximum in September. The cleansing and application of a molluscicide (copper sulphate) to the raceways in spring resulted in a 56% reduction in the numbers of metacercariae infecting trout during the following summer. However, no further improvement in parasite control was recorded when the treatment was repeated in the following year. Experiments using caged fish indicated that diplostomiasis was confined to certain areas of the farm only and that the infection rate of rainbow trout with D. Spathaceum cercariae was correlated (P < 0·01) with water temperature. The results of the study indicated that it is possible by regular cleaning and use of molluscicides to keep the intensity of diplostomiasis at such a level that rainbow trout do not become severely affected. However, as with other parasitic diseases, a combination of control methods will probably be required to eradicate the disease completely from trout farms.

Infective 3rd-stage larvae of three species of marine ascaridoid nematodes, Pseudoterranova (= Phocanema = Porrocaecum = Terranova) decipiens, Anisakis simplex and Phocascaris/Contracaecum sp. were cultured in vitro in 0·9% saline or Medium 199 at 37°C beneath an atmosphere of either air or 95% air:5% CO2 All three species responded to culture at 37°C by producing a new cuticle. The majority of P. decipiens completed ecdysis under all the culture conditions employed. Ecdysis in A. simplex was stimulated by a high concentration of CO2; this effect was particularly noticeable in saline cultures. Both A. simplex and Phocascaris/Contracaecum sp. developed more effectively in a nutrient medium than in saline.

Evidence from many sources indicates that, in several parasite–vector systems, flight ability, fecundity or gonotrophic concordance of vectors, are affected by parasites (including bacteria, rickettsia and viruses) and in some cases parasites cause well-documented pathogenic effects (see reviews, for example, by Lavoipierre (1958a) and Hawking & Worms (1961) on filaria; Molyneux (1977, 1983) for trypanosomatids; Killick-Kendrick (1979) for Leishmania).
