‘The Stockholm Pregnancy and Weight Development Study’ was conducted to identify risk factors for postpartum weight retention, such as dietary habits, physical activity and socio-demographic factors. The body weight development of 1423 pregnant women was studied prospectively from the beginning of the pregnancy until 1 year postpartum. Data were collected from routine pregnancy records and from questionnaires 6 and 12 months postpartum. Mean weight retention 1 year postpartum was 0.5 kg compared with the prepregnancy body weight. A ‘trend method’ was constructed to identify a number of pre-defined major patterns of behaviour. The weight retention 1 year postpartum was greater in women who (a) increased their energy intake during and after pregnancy, (b) increased their snack eating after pregnancy tc three or more snacks/d, and (c) decreased their lunch frequency starring during or after the pregnancy. Women who had retained > 5 kg 1 year postpartum were more seldom physically active in their leisure time throughout the study period compared with women with a smaller weight gain. Postpartum weight retention correlated negatively with the degree of physical activity in the second half year postpartum. These results indicate that postpartum weight retention is more affected by a change in lifestyle during, and above all after, pregnancy than by factors before pregnancy.

The apparent digestibility of energy, protein, fat and non-starch polysaccharides (NSP) of low and high dietary fibre (DF) mixed diets were studied in three series of experiments with man and the rat. Low DF diets were used as control diets in each experimental series and the DF level was increased by adding fruits and vegetables (Study 1), citrus fibre concentrate (Study 2) and insoluble barley fibre (Study 3), In Study 3 the high DF diet was fed at two protein levels. There was in most cases good agreement between the digestibility of energy between man and the rat, with the digestibility of energy of the low DF control diets of 0.941–0.950 in man compared with 0.933–0.952 in the rat and of the high DF diets of 0.897–0.931 in man and 0.865–0.920 in the rat. The biggest difference in digestible energy between the two species was found for the diet enriched with fruits and vegetables (0.032 absolute units) and citrus fibre concentrate (0.025 absolute units). Apparent digestibility of protein was slightly lower in man than in the rat for all diets in Studies 1 and 2. In Study 3, however, apparent digestibility of protein was consistently lower in man than in the rat with differences in absolute digestibilities between the two species varying from 0.023 (high DF/high protein) to 0.071 (high DF/low protein). The digestibility of fat was the same in man and in the rat in all but the high DF diet of Study 2. The rat appears to have a lower capacity to digest fibre polysaccharides than man and the digestibility of NSP was consistently lower in the rat than in man. The biggest difference between the two species was found for the diets in Study 2 where the digestibility of NSP in man was measured to be 0.774–0.885 compared with only 0.501–0.517 in the rat. For the other diets the differences in NSP digestibility were 0.077–0.137 absolute units. In spite of some differences between man and the rat in their ability to digest nutrients the various diets are ranked in the same order by the two species.

Six adult castrated male red deer (Cervus elaphus), fitted with rumen cannulas, were offered chaffed lucerne hay ad lib. during winter and summer, with voluntary food intake (VFI) being respectively 59 and 89 g dry matter intake (DMI)/kg bodyweight 0.75 per d. The same animals were also offered the same feed during summer, with intake restricted to that of winter VFI. The apparent digestibility of gross energy (0.60) or fibre (0.41) and the total capacity (volume) of the rumen were unaffected by season or level of intake. Relative to winter ad lib. feeding, N retention, total rumen pool size (DM +water), rumen pool size as a proportion of capacity, and rumen total volatile fatty acid (VFA) pool size were increased during summer ad lib. feeding. Relative to winter ad lib. feeding, N retention, rumen ammonia irreversible loss rate (IRL), total rumen pool size, rumen pool size as a proportion of capacity, and rumen ammonia and total VFA pool sizes were also increased during summer restricted feeding. Rumen Lignin fractional disappearance rate (FDPR) was lower in summer than in winter, and there was a nonsignificant trend for rumen fractional outflow rate (FOR) of liquid to follow the same trend. Molar proportions of acetate and propionate were unaffected by season, proportions of n-butyrate were slightly higher in summer, and proportions of iso-butyrate and iso-valerate were higher for summer restricted than for winter ad lib. feeding. When intakes were equalized there were no seasonal changes in rate of rumen water outflow, net rumen water balance or intestinal water absorption. It is concluded that there is a seasonal change in rumen physiology in red deer during summer causing increased total rumen pool size (DM + water), an increase in rumen ammonia production and pool size, and an increase in rumen total VFA pool size which are all independent of the increase in VFI. The increased total rumen pool size in the summer restricted group may indicate an increased mean retention time (MRT) of digesta in the rumen. MRT for particulate matter was calculated to be 29.2 and 34.8 h during winter and summer respectively. This, together with increased rumen ammonia production, may function to maintain rumen fibre digestion when VFI normally increases during summer. The increased rumen VFA pool size may indicate increased VFA production during summer, in the same way as ammonia IRL was increased.

The action of intravenous infusion of the βagonist cimaterol(2.5 mg/d) on whole-body N retention and protein synthesis in peripheral tissues was examined in growing sheep. Wool growth was determined from skin patch clippings and adjusted to total fibre production. Protein synthesis was measured, using sequential large dose injections of [ l-I3C]valine, leucine and phenylalanine and then [ring-d,]-phenylalanine, on biopsy samples from skin and m. longissirnus dovsi taken before β-agonist administration, at day 3 and day 15 of cimaterol infusion, and 15 d after withdrawal of the drug. Cimaterol increased total N retention by 1.9–2.3 g N/d (P < 0.01) over three successive 5 d periods. In contrast, wool growth was significantly reduced by 0.7 g N/d (P < 0.001) and the proportion of total N retained in wool declined from 0.71 to 0.25 (P < 0.01). The reduction in woo1 growth was accompanied by a decrease in protein fractional synthesis rate (FSR) in skin (11.6 v. 6.3%/d, P < 0.01). Muscle protein FSR, on the other hand, was markedly stimulated during cimaterol infusion (1.45 v. 3.01 %/d, P < 0.001) as was RNA concentration (P < 0.001), RNA:protein (P < 0.001) and protein:DNA (P < 0.05). The estimated increase in total protein synthesis in muscle (+ 24 to 30 g/d) due to cimaterol administration was counterbalanced by reductions for skin (-25 to 27 g/d); this may account for the lack of changes in whole-body protein synthesis following β-agonist administration reported in other studies. Although N retention rapidly returned to control values following withdrawal of the drug, both wool growth and skin protein synthesis remained depressed, while muscle protein FSR declined, but not to pre-treatment values. These results suggest a persistent action of cimaterol, but whether this is a function of residue concentrations or long-term metabolic responses is not known

Guanidinated proteins when fed to non-ruminants provide values for both endogenous amino acid losses and amino acid digestibilities, provided that the homoarginine residues in the treated protein are randomly distributed. Earlier studies have established that guanidination has only minor effects on the structure of the protein and, in particular, on its susceptibility to proteolysis. Furthermore, we have confirmed that homoarginine behaves as a typical amino acid in the small intestine. Lysine residues in casein and soya-bean protein, and in the proteins of cotton-seed meal, meat meal, soya-bean meal, maize, sorghum and wheat were converted to homoarginine by guanidination, the extent of conversion ranging from 37–68%. Sequential proteolysis in vitro of these guanidinated materials showed that the ratios of homoarginine to other amino acids remained unchanged for casein and soya-bean protein, indicating random distribution of homoarginine residues, but not for all the amino acids in meals and cereals. The use of guanidinated casein as the sole protein source in diets fed to broiler chickens allowed measurement of endogenous losses of amino acids under normal feeding conditions and calculation of true digestibilities of dietary amino acids at the ileum. Endogenous amino acid losses measured by the use of guanidinated casein (15.3 g/kg dry matter (DM) intake) were significantly higher (P < 0.001) than values obtained by feeding a N-free diet (5.4 g/kg DM intake), or by regression analysis to zero N intake (72 g/kg DM intake)

Two experiments were conducted to determine the utilization of ileal digestible isoleucine by growing pigs. In the first, the apparent ileal digestibility of amino acids in cottonseed meal, lupin-seed meal and soya-bean meal was determined in pigs fitted with‘T‘-shaped cannulas. In the second, three isoleucine-deficient diets were formulated to 0.23 g ileal digestible isoleucine/MJ digestible energy (DE) with the three protein concentrates contributing the only source of isoleucine in sucrose-based diets. An additional three diets were formulated with supplements of isoleucine to confirm that isoleucine was limiting in the first three diets. The growth performance and retention of isoleucine by pigs given the six diets over the 20–45 kg growth phase were then determined. The apparent ileal digestibility of isoleucine in the three protein concentrates (proportion of total) was: cottonseed meal 0.68, lupin-seed meal 0.86, soya-bean meal 0.86. There were no significant differences (P > 0.05) in growth rates (g/d) and crude protein deposition rates (g/d) of the pigs given the three diets formulated to 0.23 g ileal digestible isoleucine/MJ DE: cottonseed meal 590, 84; lupin-seed meal 613, 87; soya-bean meal 594, 91 (SEM 13.0, 2.9) respectively. The response of pigs to the addition of isoleucine confirmed that isoleucine was limiting in these diets. The proportion of ileal digestible isoleucine retained by pigs given the cottonseed meal (0.65) was slightly lower than that retained by pigs given soya-bean meal (0.73; P < 0.05). These results indicate that values for the ileal digestibility of isoleucine in protein concentrates more closely reflect the proportion of isoleucine that can be utilized by the pig than occurs for other amino acids such as lysine, threonine and methionine.

This study has investigated the influence of dietary fatty acid composition on mammary tumour incidence in N-ethyl-N-nitrosourea (ENU)-treated rats and has compared the susceptibility to dietary fatty acid modification of the membrane phospholipids phosphatidylinositol (PI) and phosphatidylethanolamine (PE) from normal and tumour tissue of rat mammary gland. The incidence of mammary tumours was significantly lower in fish oil- (29%), compared with olive oil- (75%; P < 0.04) but not maize oil-(63%; P < 0.1) fed animals. No differences in PI fatty acid composition were found in normal or tumour tissue between rats fed on maize oil, olive oil or fish oil in diets from weaning. When normal and tumour tissue PI fatty acids were compared, significantly higher amounts of stearic acid (18:0) were found in tumour than normal tissue in rats given olive oil (P < 0.05). A similar trend was found in animals fed on maize oil, although differences between normal and tumour tissue did not reach a level of statistical significance (P < 0.1). In mammary PE, maize oil-fed control animals had significantly higher levels of linoleic acid (18:2n−6) than either olive oil- or fish oil-fed animals (P < 0.05, both cases) and levels of arachidonic acid were also higher in maize oil- compared with fish oil-fed animals (P < 0.05). In tumour-bearing animals no differences in PE fatty acid composition were found between the three dietary groups. When normal and tumour tissue PE fatty acids were compared, significantly lower amounts of linoleic acid (18:2n-6; P < 0.01) and significantly greater amounts of arachidonic acid (20:4n−6; P < 0.05) were found in tumour than normal tissue of rats fed on maize oil. The present study shows that the fatty acid composition of PI from both normal and tumour tissue of the mammary gland is resistant to dietary fatty acid modification. The PE fraction is more susceptible to dietary modification and in this fraction there is evidence of increased conversion of linoleic acid to arachidonic acid in tumour compared with normal tissue. Lower tumour incidence rates in rats given fish oils may in part be due to alteration in prostanoid metabolism secondary to displacement of arachidonic acid by eicosapentaenoic acid, but PE rather than PI would appear to be the most likely locus for diet-induced alteration in prostanoid synthesis in this tissue. Effects of dietary fatty acids other than on the balance of n−6 and n−3 fatty acids, and on prostanoid metabolism, should also be considered. The significance of increased stearic acid content of PI in tumours of olive oil-fed animals and the possible influence of dietary fatty acids on the capacity for stearic acid accumulation requires further study.

The kinetics of the response in the blond of piglets to physiological oral intakes of galactose and glucose, and intravenous administration of galactose are described. Following the intravenous administration of galactose to 2- and 10-d-old piglets (n 7), the half-life was 7.98 (SD 0.75) and 7.99 (SD 1.89) min respectively, and efficient elimination rate was 9.09 (SD 2.15) and 8.75 (SD 0.79) % per min respectively. The turnover of galactose in the piglets was 100-3 μg/min per kg body weight. These observations demonstrate that galactose was rapidly removed from the blood of the piglets. While the dosing and sampling procedures stimulated hyperglycaemia, they had no effect on the concentration of galactose in the peripheral plasma. The galactose area under the curve (adjusted to the plasma volume of the animal) following a dose of either galactose or galactose plus glucose was 1.75 (SD 0.15) and 1.95 (SD 0.14) arbitrary units respectively in 2-d-old piglets and 1.96 (SD 0.26) and 1.98 (SD 0.10) arbitrary units respectively in 10-d-old piglets. Since the presence of glucose did not lower the adjusted area under the curve for galactose in the peripheral blood, the effect of glucose on the metabolism of galactose in piglets was more like that reported for rats than that for man, guinea-pigs or mice. It is suggested that the galactose moiety of lactose may make an important contribution to the replenishment of liver glycogen in the neonatal piglet.

The postprandial glycaemic response following a meal is reduced with the addition of soluble dietary fibre. The reductions in the glycaemia are thought to be due largely to increased viscosity of the gastrointestinal (GI) contents retarding digestion and absorption. The aims of the present study were to determine the effect that the GI tract has on the viscosity of meals containing different soluble fibres and to determine whether the glycaemic response of a meal (containing the soluble fibre) was predicted by the viscosity of the digesta in the small intestine. High carbohydrate diets containing 70 g soluble fibre (guar gum, xanthan gum or methylcellulose)/kg or 70 g insoluble fibre (wheat bran)/kg were diluted in water to a final fibre concentration of 18 g/kg. Following dilution the wheat bran diet had no measurable viscosity, while the viscosities of the soluble fibre diets were elevated. When the diets were fed to male Sprague–Dawley rats for 2 weeks the viscosities of the stomach and small intestinal digesta were not predicted by the viscosity of the diets measured before ingestion The action of the GI tract on the viscosity of the soluble fibres was investigated in vitro by dilution of the diets with acidic and neutralizing solutions, mimicking gastric and duodenal secretions. Dilution of diets with either acidic and neutralizing solutions or saline control significantly lowered the viscosity of all diets, while alterations in the pH of the diets had little impact on the resultant viscosity. When fasted rats were orally administered with the differing diets (0.25 g carbohydrate/kg body weight), the postprandial glucose response was reduced following the soluble-fibre-containing meals when compared with the wheat bran-supplemented meal, although the reduction in glycaemia only reached statistical significance with xanthan supplementation. These results indicate that there are large changes in the viscosity of a meal containing soluble fibre following ingestion, and that dilution of the diet by GI secretions is important in determining the resultant viscosity in the small intestine. Furthermore, the large differences in viscosity of the GI contents following consumption of the diets containing the soluble fibres were not predictive of the postprandial glycaemic response.

Cryptomys hottentotus has no access to dietary or endogenous sources of cholecalciferol (D3). Serum concentrations of calcifediol (25(OH)D3) were undetectable (< 5 ng/ml) and calcitriol (1,25(OH)2D3), although detectable in plasma, was at a low concentration (31.40) (SEM 3.06 pg/ml). Despite their naturally impoverished vitamin D status, these animals exhibited highly efficient mineral absorption processes (Ca apparent fractional absorption efficiency, AFA (%) 95.33 (SEM 1.36); inorganic P (Pi) AFA (%) 93.49 (SEM 0.81)). Furthermore, plasma mineral content was tightly regulated (Ca 2.57 (SEM 0.08) mmol/l; Mg 1.23 (SEM 0.05) mmol/l; Pi 2.12 (SEM 0.15) mmol/1). Mode of uptake in C. hottentotus was unlike that in other D3-replete mammals. First, passive (rather than active) uptake occurred in the traditional site of active Ca absorption (with serosal: mucosal (S:M) ratios in the duodenum of 1.32 (SEM 0.13)), and the only site of active Ca uptake was the hindgut (caecum S:M 3.35 (SEM 0.46); proximal colon S:M 2.13 (SEM 0.30)). Despite the presence of active uptake in these hindgut regions, their overall contribution to the daily rate of mineral absorption was small (9.53 (SEM 1.27)%). These underground inhabitants rely upon highly efficient, passive mineral uptake. This is adequate to meet their mineral requirements and maintain mineral homeostasis in the absence of vitamin D.

The effects of dietary protein on Ca bioavailability and utilization in bone were examined in male spontaneously hypertensive rats (SHR) fed on diets containing either casein (200 g/kg (control), 60 g/kg or heat-damaged (HD) 200 g/kg) or soya-bean protein isolate (200 g/kg (control), 60 g/kg, or HD 200 g/kg). Casein was heat-damaged to limit caseinophosphopeptide (CPP) production in order to evaluate casein enhancement of Ca bioavailability. All diets contained an adequate level of Ca (5 g/kg). A 24 h mineral balance study was performed when animals were 10 weeks old, followed by measurement of in situ paracellular Ca disappearance, femur mineralization and biomechanics at 14 weeks of age. Digestibility of soya-bean and both HD proteins estimated in vitro was reduced compared with native casein. Animals fed on HD and 60 g/kg protein diets exhibited decreased (P < 0.05) body weight gain, dry matter intake and feed efficiency compared with controls. The ileal disappearance of 45Ca was lower (P < 0.05) in animals fed on HD casein and all the soya-bean protein diets. Ca balance was not strongly affected by dietary treatments. A significant (P < 0.05) interaction between protein source and reduced protein intake was observed for femur calcification and physical measurements. Femur bending failure energy and biomechanical force measurements were reduced (P < 0.05) in HD and 60 g/kg casein and soya-bean protein fed animals. These findings suggest that whole-body Ca homeostatic mechanisms were involved in compensating for reduced Ca bioavailability and retention from casein diets modified to reduce protein digestibility and CPP production.

Bioefficiencies of α-, γ- and δ-tocopherol in comparison with all-rac-α-tocopherol were established in broiler chickens. For this, 1-d-old male broiler chickens received a diet deficient in vitamin E and supplemented with increasing doses of the corresponding tocopheryl acetates. After 2 and 3 weeks of feeding, the animals were killed to obtain blood and liver samples. The ex vivo tests used were detergent induced haemolysis and pentane production by liver microsomes. Bioefficiencies were calculated by comparison of the dose–response curves. It is concluded that haemolysis and pentane production are appropriate indicators of the bioefficiency of tocopherols in broiler chickens. The values obtained by both tests hardly differed and agree well with the figures previously obtained from rats and other species.

The antibody response to a dietary antigen (soya-bean protein) and the development of oral tolerance was studied in pigs in a family pen system where the piglets are left with their mothers and gradually wean themselves onto a soya-bean-based diet over a 12 week period. In the first experiment three groups of pigs (eight pigs/group) aged either 2, 8 or 13 weeks were immunized with soya-bean protein or ovalbumin (OvA; controls) intra-peritoneally (i.p.) in Quill A adjuvant and subsequently boosted 2 weeks later. All groups showed an IgG response to the injected antigens indicating lack of tolerance induction to the dietary antigen. Interestingly the groups injected with OvA showed an almost identical response to soya- bean protein as the groups injected with soya-bean protein. In a second experiment with a similar protocol, soya bean was withdrawn from the feed before immunization which resulted in lack of response to soya-bean protein in the groups injected with OvA and a lack of response to injected soya-bean protein in the 14-week-old group, indicating that systemic tolerance was established by 12 weeks of age. The results from the two experiments suggest a compartmentalized response to soya-bean protein i.e. local antibody production to dietary soya bean along with systemic tolerance to injected soya-bean protein. The work also suggests that delayed‘natural’weaning may avoid damaging hypersensitivity reactions associated with early weaning.

The effects of condensed tannins in hulls of faba beans (Vicia faba L.) on the activities of trypsin (EC3.4.21.4) and chymotrypsin (EC3.4.21.1) in digesta obtained from the small intestine of pigs were studied. Using four castrated male pigs (mean body weight 83 kg) fitted with both a simple T-cannuIa in the duodenum and a post-valvular T-cannula at the terminal ileum, two experimental diets were tested in a Latin square design. The low-tannin diet (LT) contained 200 g faba bean hulls (cv. Blandine)/kg with a low content of condensed tannins (< 0.1 % catechin equivalents). The high-tannin diet (HT) contained 200 g faba bean hulls (cv. Alfred)/kg with a content of condensed tannins of 3.5 % catechin equivalents. Spot samples of fresh duodenal digesta were taken daily at fifteen time points between 08.00 and 20.00 hours on four consecutive days. Ileal digesta were collected nearly quantitatively on the same days between 08.00 and 20.00 hours over periods of 2 h. Trypsin and chymotrypsin activities in duodenal digesta did not differ between treatments at any time point (P > 0.05). In ileal digesta of pigs given diet HT the mean activity of trypsin was reduced (P < 0.05). The activity of chymotrypsin in ileal digesta did not differ between treatments. Trypsin activity: chymotrypsin activity was somewhat lower in ileal digesta of pigs receiving the HT diet (P < 0.10). The apparent ileal digestibility of crude protein (N × 6.25) was lower for the HT than for the LT diet (0.614 v. 0.728; P < 0.05). Condensed tannins are probably responsible for the lower activity of trypsin in ileal digesta of pigs fed on high-tannin faba bean hulls. Various explanations for the absence of effects of condensed tannins on enzyme activity in duodenal digesta are discussed.
